Abstract
Objective: To investigate the effects of resveratrol and rutin on the secretion of interleukin-6 (IL-6) and interleukin-8 (IL-8) from human mononuclear cells (MNCs) and polymorphonuclear cells (PMNCs) following the stimulation with lipopolysaccharide (LPS) or peptidoglycan (PGN), and to understand the clinical implications in the treatment of sepsis. Methods: Whole blood samples were obtained from 10 healthy volunteers. White cells were harvested and treated with 20 mg · L-1 LPS or 20 μmol · L-1 PGN based on the IC50. After 4-h incubation at 37°C and 5% CO2, 20 μmol · L-1 resveratrol or 50 μmol · L-1 rutin were added into the culture media. IL-8 and IL-6 concentrations in conditioned culture media were determined by the methods of ELISA 24 h later, and the values in positive and negative control groups were compared. Results: IL-8 secretion from MNCs stimulated with LPS and IL-6 secretion from PMNCs stimulated with PGN were significantly decreased by both resveratrol and rutin (for IL-8, resveratrol: P < 0.01, rutin: P < 0.05, n = 10; for IL-6, resveratrol: P < 0.05, rutin: P < 0.05, n = 10). Conclusion: Resveratrol and rutin may exert different inhibitory effects on the cytokine secretion of human MNCs and PMNCs.
