This chapter explores how various foodomics technologies based on liquid chromatography--mass spectrometry?mass spectrometry (LC--MS?MS) are used to examine mechanistically the links between folate status and human health. The importance (and difficulties) of accurately measuring blood folates using LC--MS?MS in human population monitoring is discussed. The application of proteomics to investigate the effect of folate status on global protein expression and metabolic pathways associated with immune function, genomic stability, and malignant transformation is described in detail using examples from cell and animal models and from a human intervention study. Finally, the use of LC--MS?MS to investigate the impact of folate status on abnormal DNA methylation as a common mechanism linking altered gene expression and cell proliferation in both colon cancer and CVD is discussed.
